GLI1与胃癌上皮间质转换的关系及临床研究(1)

     [摘要]目的 观察GLI1表达与胃癌上皮间质转换的关系。方法 收集2006年1月~2011年12月诊断明确、资料完整的58例手术切除胃癌标本,做成组织芯片,采用免疫组化SP的方法检测组织芯片中胃癌组织及正常胃黏膜组织中GLI1、E-cadherin、N-cadherin的表达。结果 GLI1在胃癌中表達较正常组织高,与胃癌的淋巴转移、临床分期、Lauren分型、肿瘤分化明显有关,差异有统计学意义(P<0.05);E-cadherin在正常胃黏膜组织表达比胃癌组织高,与胃癌的淋巴转移、临床分期、Lauren分型、肿瘤分化明显有关,差异有统计学意义(P<0.05);N-cadherin在胃癌中表达高于正常胃黏膜组织,与胃癌的淋巴转移、临床分期、Lauren分型、肿瘤分化明显有关,差异有统计学意义(P<0.05);GLI1表达与上皮间质转换的状态有关,差异有统计学意义(P<0.01)。结论 在胃癌组织中GLI1表达与E-cadherin、N-cadherin有关,提示GLI1可能参与了EMT过程。

    [Abstract]Objective To observe the relationship between GLI1 expression and epithelial-mesenchymal transition in gastric cancer.Methods 58 cases of gastric cancer specimens who were given surgical resection with clear diagnosis and complete data from January 2006 to December 2011 were collected,and tissue microarray was made.The expression of GLI1,E-cadherin and N-cadherin in gastric carcinoma and normal gastric mucosa in tissue microarray were detected by immunohistochemical SP method.Results GLI1 expression in gastric cancer was higher than that in normal tissues (P<0.05),which was significantly associated with lymph node metastasis,clinical staging,Lauren typing and tumor differentiation of gastric cancer,with significant difference (P<0.05).The expression of E-cadherin in normal gastric mucosa was higher than that in gastric cancer (P<0.05),which was significantly associated with lymph node metastasis,clinical staging,Lauren typing and tumor differentiation of gastric cancer,with significant difference (P<0.05).The expression of N-cadherin in gastric cancer was higher than that in normal gastric mucosa (P<0.05),which was significantly associated with lymph node metastasis,clinical staging,Lauren typing and tumor differentiation of gastric cancer,with significant difference (P<0.05).GLI1 expression was associated with epithelial-mesenchymal transition,with significant difference (P<0.01).Conclusion The expression of GLI1 in gastric cancer is associated with E-cadherin and N-cadherin,indicating that GLI1 may be involved in the EMT process.

    胃癌(gastric cancer)是最常见的恶性肿瘤之一,发病率在全球常见癌症中居第4位,在癌症相关死因中居第2位,每年因胃癌死亡者约70万例[1];其在我国常见癌症中居第2位,在癌症死因中居第3位[2]。我国中晚期胃癌患者术后5年的生存率为20%~30%,早期胃癌患者的生存率最高为85%~100%[3-4]。决定患者生存率的主要因素是转移,因此寻找肿瘤转移的标志物是临床研究的方向。研究发现上皮间质转换(epithelia-mesenchymal transition,EMT)现象及SHH信号通路中的GLI1表达与肿瘤的转移密切相关。EMT是一种基本的生理病理现象,其表达上调的因子主要是N-cadherin,表达下调的因子主要是E-cadherin。本研究以胃癌标本作为研究对象,采用免疫组化检测胃癌中SHH信号通路中核转录因子GLI1及E-cadherin、N-cadherin的表达情况,现报道如下。, http://www.100md.com(林智平 段炤)
