化痰祛瘀汤对失眠大鼠模型的治疗作用及机制(1)
摘要 目的：觀察化瘀祛痰汤对失眠大鼠模型的治疗作用，探讨作用机制。方法：选取40只成年SD大鼠，分为正常对照组、模型对照组、中药观察组和艾司唑仑组。建立对照组之外3组大鼠失眠模型。中药观察组给予化瘀祛痰汤，艾司唑仑组给予艾司唑仑水溶液灌胃，模型对照组和正常对照组均给予同等体积的蒸馏水灌胃。观察各组建模及治疗后行为学变化;比较各组大鼠干预前后自主活动数;比较干预后各组大鼠戊巴比妥钠致眠后睡眠潜伏期和睡眠持续时间;比较各组大鼠下丘脑5-HT和5-HIAA水平，下丘脑GABA蛋白表达。结果：中药观察组大鼠治疗后的行为学改善、自主活动数稳、戊巴比妥钠致眠后睡眠潜伏期和睡眠持续时间、下丘脑5-HT和5-HIAA水平、下丘脑GABA蛋白表达改善均优于艾司唑仑组大鼠及模型对照组大鼠，差异有统计学意义( P <0.05)。结论：化瘀祛痰汤对失眠大鼠有良好的治疗作用，能够改善行为学和自主活动，延长睡眠潜伏期和睡眠持续时间，效果明显优于艾司唑仑。

Abstract Objective： To observe the therapeutic effects of Huayu Qutan Decoction on insomnia rats and explore its mechanisms. Methods： A total of 40 adult SD rats were divided into a normal control group，a model control group，a traditional Chinese medicine treatment group and an estazolam group.The insomnia model was established in 3 groups except for the control group.Traditional Chinese medicine treatment group was given Huayu Qutan Decoction，estazolam group was given estazolam solution gavage，the model control group and the normal control group were given the same volume of distilled water gavage.The behavioral changes of rats in each group after modeling and treatment were observed; the number of autonomous activities of rats in each group before and after intervention were compared; the sleep latency and duration of sleep induced by pentobarbital sodium in each group after intervention were compared; and the content of 5-HT and 5-HIAA in hypothalamus and the expression of GABA protein in hypothalamus in each group were compared. Results： The improvement of behavior，number of autonomous activities，sleep latency and sleep duration induced by pentobarbital sodium，5-HT and 5-HIAA contents in hypothalamus，and GABA protein expression in hypothalamus in the traditonal Chinese medicine treatment group were better than those in the estazolam group and the model control group ( P <0.05 ). Conclusion： Huayu Qutan Decoction has good therapeutic effects on insomnia rats.It can improve behavior and autonomic activity，prolong sleep latency and sleep duration.The effects are better than that of estazolam.It is speculated that it is related to the significant increase of 5-HT and 5-HIAA contents in hypothalamus and the promotion of GABA protein expression., http://www.100md.com(王琪 李延 李勇 吴限)
